The Hard Coating of the Spores
Spores are highly complex cell structures that have evolved as survival strategies in response to harsh environmental conditions, and provide energy-efficient surface area for growth. Spores take the shape of oval or rod-like spores with thick cell walls (called an endosporium), as well as an external proteinaceous coat known as an exosporium. Their oval/rod form makes them great at withstanding immense internal pressure generated during germination processes (>7.0 Mpa). Their circular form maximizes surface area for further growth – ideal for protecting cells against environmental stressors such as pressure generated during germination processes.
Additionally, the spore core is made up of an insoluble, heat-stable gel believed to protect spores from water molecules. Finally, its membrane is protected by an extracellular matrix composed of complex glycoprotein called Bacillus collagen-like protein A (BclA), which may serve as an immunodecoy and prevent interactions between spores and host cells in their environment.
Outside this outer layer, spores contain numerous enzymes that inactivate H2O2, including three major superoxide dismutases (KatA, KatB and KatX), DNA-protective MrgA protein and alkylhydroperoxide reductase Ahp (all identified). Furthermore, H2O2 could potentially be actively destroyed by catalases present within their coat layers as mutations did not alter resistance to lethal effects of H2O2.
Spores are shielded from environmental radiation by inner and outer spore coat layers, as previously discussed. While chemical or genetic methods to remove them did not alter resistance to 254-nm UV-C radiation; mutant strains AD17 and AD142 that lack outer spore coat due to gerE36 cotE::cat mutation were more sensitive to solar UV-C radiation.
Spores from mutant spore coat strains were more sensitive to UV-B radiation’s sporicidal effect than those from wild-type PY79 strain, further supporting that the inner and outer spore coats are essential for resisting environmental UV wavelengths. Indeed, it takes over 1,000kg of Ganoderma lucidum spores in order to yield one kg of Organo Gold Spore Powder!
The Cracking of the Spores
Spores of Ganoderma mushrooms are at the core of ORGANO’s King Coffee and other products designed to promote natural vitality, and ORGANO scientists in China have spent years learning how to harvest spores without harming their precious content. Unfortunately, however, harvesting them is no simple task; their hard outer shell protects them from human bodies while an inner layer prevents their nutrients from penetrating them fully. Scientists at ORGANO China have spent time cracking this shell without risking its contents being lost within. At the end of their one year life cycle, when our spores have reached maturity they resemble fine baby powder and can be collected manually with special brushes. It takes 1,000 kilograms of these powdery spores to create one kg of our product-grade spore powder; therefore, each single speck of these spores is highly valued and treated as such.
To protect the spores from environmental stressors and ensure that they remained viable during harvesting, we encased them in a 2% (w/v) solution of sodium alginate in ethanol for 24 h before freeze drying (Christ Alpha 2-4/LD Plus, Germany). This technique enabled successful preservation while simultaneously showing high metabolic activity during freeze drying.
This technique may provide an effective microencapsulation approach for preserving Bacillus sphaericus spores in self-healing concrete, with superior survival rate and yield compared with spray and freeze drying methods. Furthermore, this study highlights the necessity of understanding bacterial spore physiology so as to create more efficient techniques for their preservation.
