We conducted a systematic review of RCTs of Ganoderma lucidum supplementation studies involving human participants and health outcomes, with subgroup analyses performed according to health conditions, dosage, intervention duration duration sample size and country.

Subgroup analyses performed using health condition (chronic disease vs. at-risk but generally healthy populations), Ganoderma dose (1400 mg/day vs. > 1400 mg/day), Ganoderma dosage duration (1-24 weeks), age of participants 50 years vs > 50 years; geographical region (East Asia vs Middle East vs America, Oceania and Europe); sample size 40 participants or >40 participants) revealed Ganoderma supplementation as having positive impacts on BMI, creatinine, ALT and MDA levels; but their effects did not differ by geographical region or age.

To address these concerns, we conducted an exhaustive systematic review and dose-response meta-analysis of RCTs of Ganoderma lucidum on health outcomes. To be included, RCTs must compare Ganoderma lucidum against control interventions while reporting health-related outcomes; over 4224 articles were obtained from electronic databases including MEDLINE (via PubMed), Cochrane Library, Scopus and Web of Science without restrictions for publication language or participant age as of August 2024, while all bibliographies of all articles were reviewed to identify additional relevant references for further research purposes.

Results indicate that the pooled estimate for intervention effects on creatinine is an estimated decrease by 95% confidence interval from -0.554 to 1.606; I2 = 80%). By contrast, estimates for SBP and TC do not differ significantly across groups due to high heterogeneity among trials due to differences in study populations, dosage and intervention protocols or geographical regions.
